Orla technology allows the immobilisation of proteins to surfaces in a highly ordered and functional manner. The protein of interest is fused to a proprietary surface-binding protein to produce a fusion protein. This can bind to surfaces anchored via the surface binding protein leaving the protein of interest distal to the surface, structurally intact and with high retention of function.
This technology is offered for use in a variety of applications:
1. Protein immobilization for biosensor chip functionalization. Immobilization of antigens, small epitopes, and single-chain antibody fragments as capture surfaces. Proven to have higher surface exposure compared to adsorption or chemical coupling.
2. Protein immobilisation on ELISA plate surfaces. Immobilization of antigens, small epitopes, and single-chain antibody fragments as capture surfaces. Proven to have higher surface exposure compared to adsorption or chemical coupling.
3. Coated plates for cell culture. Surfaces coated with ECM proteins and/or growth factors (see www.orlaproteins.com for a full list of available proteins). Highly reproducible. Try our OrlaExplorerTM plates to discover the optimal growth surface for your cell/media combination. The coating is transferable across a range of formats – multi-well plates, flasks, 3D matrices, beads etc. Uniquely, we can mix and match different ECM motifs and growth factors on the same growth surface to closely mimic in vivo conditions.
4. Immobilisation of enzymes. The use of an anchor protein allows us to design and engineer the fusions in such a way as to control the position and orientation of the enzymatic domains and active sites, thereby retaining higher structural integrity compared to direct adsorption or chemical coupling.
In addition, we provide services for the contract manufacturing of recombinant proteins at the 1 mg-100 mg scale. Ideal for antigen production, pilot experiments etc. We express in E. coli either as soluble protein or, if necessary, as inclusion bodies and have a wealth of experience in the refolding of proteins from inclusion bodies.
